Développement de biocapteurs à base de réseaux de nanofils de silicium aléatoirement orientés pour une détection électrochimique ou à effet de champ

Résumé

Randomly oriented nanowire networks (nanonets) exhibit interesting properties, arising from the intrinsic properties of the individual nanostructures, but also offer remarkable new properties related to the nanonet structure, namely a better reproducibility, and high fault tolerance and flexibility. In this context, this project investigated the possibility of using a 2D assembly of randomly oriented Si nanowires (Si nanonet), as building blocks for the fabrication and development of two different types of biosensors: Si nanonet-based FET biosensors for the electrical detection of DNA hybridization, and Si nanonet-based electrochemical biosensors for thrombin detection.For this purpose, the development of each type of sensor was divided into three parts: (i) fabrication of the Si nanonet-based device, (ii) biofunctionalization of the device allowing the grafting of the bioreceptor (TBA-15 aptamer), and (iii) detection of the target molecule.Regarding the Si nanonet-based FET biosensors, the devices were fabricated following a process previously developed at LMGP. Here, the work focused on the implementation and the optimization of a biofunctionalization protocol, enabling the proper immobilization of the bioreceptor on the surface of the Si nanonet-based FETs. Then, the ability to electrically detect the DNA hybridization in dry environment was studied. Concerning the electrochemical biosensors, it was developed for the first time a process allowing the fabrication and biofunctionalization of a 2D Si nanonet-based biosensor, enabling thrombin detection in nM concentrations, indicating either the risk or the presence of thrombosis.These promising results open the door to future projects implying the use of 2D Si nanonet-based devices in the field of biodetection.
Les réseaux de nanofils aléatoirement orientés (nanonets) présentent des propriétés intéressantes, liées aux propriétés intrinsèques des nanostructures individuelles, mais offrent également de nouvelles propriétés remarquables associées à la structure du réseau, à savoir, une bonne reproductibilité, une grande tolérance aux défauts et une flexibilité importante. Dans ce contexte, ce projet a étudié la possibilité d'utiliser un assemblage 2D de nanofils de Si aléatoirement orientées (Si nanonet), comme élément de base pour la fabrication et le développement de deux types différents de biocapteurs: des biocapteurs à transistor à effet de champ (FET) pour la détection électrique de l'hybridation de l'ADN, et des biocapteurs électrochimiques pour la détection de la thrombine.À cette fin, le développement de chacun de ces biocapteurs a été divisé en trois parties : (i) fabrication d'un dispositif à base de nanonets de silicium, (ii) biofonctionnalisation du dispositif permettant le greffage du biorécepteur (TBA-15 aptamère) et (iii) détection de la molécule cible.En ce qui concerne les biocapteurs FET à base de nanonets de silicium, les dispositifs ont été fabriqués selon une méthode précédemment développée au LMGP. Ici, le travail effectué s'est concentré sur la mise au point et l'optimisation d'un protocole de biofonctionnalisation, permettant l’immobilisation correcte du biorécepteur sur la surface du dispositif FET. Puis, la capacité des biocapteurs FET à détecter électriquement l'hybridation de l'ADN en milieu sec a été étudiée. Concernant les biocapteurs électrochimiques à base de nanonets de silicium, il a été développé pour la première fois une procédure pour la fabrication et la biofonctionnalisation d'un biocapteur électrochimique à base de nanofils de silicium 2D, permettant la détection de la thrombine dans des concentrations de l’ordre du nM, indiquant le risque ou la présence d'une thrombose.Ces résultats prometteurs ouvrent la porte à de futurs projets impliquant l'utilisation de dispositifs basés sur les nanonets de silicium dans le domaine de la biodetection.
