Development of HPLC/UV and HPLC/MS analytical methods for FR-900098 : a novel antimalarial compound experimental pharmacokinetic studies on rat and micro-pig

FR-900098, un dérivé proche de la fosmydomicine, inhibite la croissance du parasite Plasmodium falciparum de manière deux fois plus efficace. Une méthode analytique basée sur la chromatographie ionique a été développée en vue de la détermination des paramètres pharmacocinétiques de FR-900098 dans le plasma humain, le plasma de rat, de micro-porc et dans la matrice d'urine de rat. La séparation de FR-900098 s'est faite sur une colonne AS11-HC (250 mm x 2 mm, Dionex), avec un temps de rétention tr =12. 25 min ± 0. 2 min et une détection UV à 230 nm. La phase mobile a consisté en une solution d'hydroxyde de sodium (pH 12. 5) traversant la colonne chromatographique à un flux de 0. 25 mL/min. L'analyse s'est déroulée à une température de 25°C. Un couplage Chromatographie Ionique-Spectrométrie de Masse a été fait. Quand bien même la sélectivité fut améliorée, la sensibilité de la méthode CI-SM est restée identique à celle obtenue avec une détection UV. Deplus, sachant que ce couplage prolonge le “run” de 40minutes, il a été decidé d'y renoncer au profit de la détection UV. Cette méthode CI-UV a permis le descriptif des paramètres pharmacocinétiques de FR-900098 chez le rat et le micro-porc après administration orale et intravéneuse. L'administration d'une dose unique de 27mg/kg de FR-900098 (équivalent free acid) présente, huit heures après administration, des concentrations plasmatiques supérieures aux IC50 et IC90 de la molécule. Afin d'améliorer la biodisponibilité orale de FR-900098 sel de mono-sodium, nous avons travaillé avec FR-900098 sous forme de sel de trométamol. A dosages équivalents et en comparaison au sel de mono-sodium, FR-900098 sel de trométamol a permi une nette augmentation des concentrations plasmatiques avec un facteur deux. La biodisponibilité orale est restée identique à celle obtenue en travaillant avec FR-900098 sel de mono-sodium.

An analytical method based on ion chromatography was developped for the determination of pharmacokinetic parameters of FR-900098 in human plasma, rat plasma, micro-pig plasma, and rat urine. FR-900098 was separated on a AS11-HC column (250 mm x 2 mm, Dionex), with a retention time of tr =12. 25 min ± 0. 2 min. Ultraviolet detection was set at 230 nm. The mobile phase consisited of a solution of sodiumhydroxyd (pH 12. 5), running through the column at a flow rate of 0. 25 mL/min. The chromatographic analysis was operated at 25°C. We tried to improve the method by a coupling with a mass spectrometer. Even if the selectivity was higher, the sensitivity of the IC-MS method was identical to the IC method. Knowing this coupling needs 40 minutes more compare to the single IC method, we decided to renounced to the mass spectrometry. From the IC method, pharmacokinetic parameters of FR-900098 were described after oral and intravenous administration in rats and micro-pigs for the first time. Eight hours after administration of a single dose of 27 mg/kg FR-900098 equivalent free acid, the plasma level was higher to the IC50 and to the IC90 of the molecule. A test with FR-900098 trometamol salt form was done in order to improve the oral bioavailability. Although FR-900098 trometamol salt form improved the plasma levels after oral or intravenous administration with a factor two, the bioavailability was not higher compare to the FR-900098 mono-sodium salt form.
